Purpose and project planning The aim of the BioBarkOak project is assessment valorization opportunities of oak bark from Romania, for the obtainment of high-tech-value bio-products and their functionalization in biomedical applications. The issues that represented the premises of the project refer to: (1) a large amount of forest waste; (2) the lack of high valorization of the biomolecules contained in waste bark; (3) alarming reports of growing microbial resistance causing different severe infections. Analyzing the identified problems, we hypothesize that the bark of woody plants can be an important source of bioactive compounds with high potential for recovery and functionalization. Thus, the I-st objective consists in a comparative study of bioactive compounds identification in the bark of the different oak species with frequent distribution in the Romanian forests, being used as raw material in furniture manufacturing. In the II-nd objective, we intend to assess the bioactivity of oak bark extracts (BOQE) and compared their antibacterial and antifungal effects. Also, the obtained results can demonstrate the ability of BOQE to potentiate the antimicrobial effect of gentamicin, to prevent biofilm formation or to influence the adhesion of some bacteria on some medical devices (mono/multifilament suture materials, venous/urinary catheters). The results regarding the permeabilization of the bacterial membrane and the alteration of the bacterial DNA can allow the evaluation of the way and the degree to which BOQE acts on the bacterial cell. The III-rd objective will be the assessment of the in vitro enzymatic properties of BOQE. In this context, our project is relevant and has a high degree of novelty by aiming to assess the multifunctional activities of BOQE and its potential in biomedical uses. By the end of the project, we will provide a valid, reliable and reproducible experimental proof concept about BOQE.

Results – the bioactive compounds identified in different Quercus bark, from Romania;

– the obtaining of natural products with antibacterial and antifungal activity and which can potentiate the antimicrobial effect of gentamicin;

– the obtaining of natural products which prevent biofilm formation or influence the adhesion of some bacteria on medical devices;

– contributions allowing elucidation of the mechanism of natural biomolecules at the cellular level;

– the obtaining of natural products with antidiabetic, antitumoral, or neuroprotective potential.
